A Scalable Engineered Extracellular Matrix Platform to Expand Tumor Cells

The demand for high-throughput and scalable cell expansion platforms that can accommodate diverse types remains a critical requirement across various biomedical fields. Fibronectin (Fn), an essential component of the extracellular matrix (ECM), has been used as conformal surface coating two-dimensional (2D) culture systems. However, soluble, globular Fn 2D coatings differs structurally from native Fn, which possesses three-dimensional (3D) fibrillar structure. Herein, large-scale engineered ECM (EECM) platform based on 3D network spanning over centimeters is presented. Extended networks are formed by shearing dilute solutions tessellated polymeric scaffolds, conveniently prepared printing. structure size Fn-based EECM scaffold optimized evaluating proliferation colorectal tumor line, CT26, commonly in vivo immunotherapy models. scaffolds support fourfold more efficient than conventional system, demonstrating potential efficacy supporting robust cancer cells ex with eye immunotherapy.
